Can you block the out from the pump and rear of the intake or just run a hose between them ? Going to start a engine on a run stand Car probably won't have a heater also.

Factory heater delete cars simply blocked the ports. There is no need to run a hose. Consider that there is no hose going to the rear driver side anyway, so what makes the passenger side different?
For a quick job you can just throw rubber caps on them. Every rubber cap I've used in the past few years has failed within a few months, so keep an eye on them.
Next time you have the water pump off the nipple can be tapped for a plug and you can replace the heater control valve with a plug.
Of what? The extra port on the water pump got a rubber cap with a wire spring hose clamp. The nipple at the back of the intake was replaced with a pipe plug.
Someone out there must have a 'real' heater delete car and pics of the fittings.
That's exactly what the factory shows in the Assembly Manual drawings. Whether that particular car is real or not I cannot say, but the cap is correct, as is the wire spring clamp.
