Congrats on the car. First off your car is a fastback model not the supreme model. Secondly, I cannot give you any advice on moving forward with your project but there sure will be people on this forum that can help you out. Great luck going forward.

No it doesn't, not sure what "S" stands for, maybe Salon, Standard? IDK.

Also that doesn't appear to be original 350 due to the fact it is blue in color and has an EGR sys.and HEI ignition. Original 350 would have been gold and have points. Be careful choosing cam because you have a brake booster that will require some vacuum to function.
Congrats on car though, looks to be fairly intact. Whats interior and rest of body like?

Welcome. The "S" stands for "S", period. It's a made up marketing name. Don't read anything into it. More to the point, the VIN will tell you exactly what you have. A 1970 Supreme hardtop has a VIN that starts 342570... Yours does not. All 1970 Supreme two door hardtops were the notchback body only. Of course, the differences are only skin deep. The chassis, suspension, and drivetrain are the same. FYI, this is a 1970 Supreme:

Also, be advised that the EGR valve and corporate blue paint tells me that is not the original motor in your car. Apparently that's a later Olds 350, probably from the mid-70s.

If your car is an automatic, you can’t just put in a 4 speed. First of all is the crankshaft would have to machined for the necessary pilot bearing a 4 speed would need. Plus you need all the lineage and pedals etc. welcome and good luck on whatever you decide to do.

Possibly because the engine in it now didn't have a crank drilled for a manual.

That’s what I was thinking also.. and have you had the car running ? It may have a decent cam in it already since it has slicks ...
