Not difficult, but can be time consuming and challenging to realign the window when reinstalling. The motor is an assembly component with the regulator, so has to be removed as an assembly after disassembling the glass from the regulator. Beware the power connector to the motor can be practically seized to the connectors on the back of the switch and on the motor. Be careful not to yank the wires out of the connectors or to break the connectors. It is VERY helpful to make witness marks for every nut and bolt on the inner panel before you take anything loose or out (ask me how I know
). There are a couple videos on Youtube explaining/demonstrating. The one I found most helpful was for a '68 or '69 chevelle - it is exactly the same under the trim panel.

As mentioned mark everything, take pictures and measurements of where everything is before disassembly.
Note how good it currently seals and aligns with the roof and front door. Note specific hardware and regulator stop locations.
Does the outer sweep need to be replaced? If so buy the set, (MAWs). Nows the time to replace it and any worn weather seals. Much easier with the glass out of the way.
Obviously have the rear interior panel and seat removed.
Reinstall the motor and regulator assembly and see how it fits without touching the front glass or top rail. If its ok you win. If not the door glass will need adjustment. Its a labor of patience with window adjustment.
TIP 1. Make sure the door aligns well with the body and the door hinge pins are not sloppy. Fix the hinges and body alignment before touching the glass or you'll be chasing your tail.
TIP 2. If you have to go at the door glass it helps to remove the front seat. This gives you good access to play with the whole side and adjust it as a unit.
TIP 3. Have a safe place to lay down the glass. Breakage sux!
